Macmahon is part of a joint venture awarded the $812 million contract to build the South Road Superway connecting the Port River Expressway to Regency Road, north of Adelaide.
The joint venture includes John Holland and South Australian company Leed Civil & Engineering.
Macmahon Chief Executive Officer Nick Bowen said the contract award demonstrates Macmahon's capability in the area of road construction.
"This is an exciting project for the Construction Business and it is a pleasing addition to our many existing road construction contracts around Australia.
"The combined expertise and capability of the joint venture partners provides an excellent platform todeliver a successful outcome for all stakeholders involved in this innovative project.
The project involves the construction of a new 4.8 kilometre highway corridor, including 2.8 kilometres of elevated roadway, with construction to start immediately and completion due in 2013.

Macmahon Holdings Limited (ASX:MAH) as part of the Urban Superway Joint Venture, has been awarded the North - South Transport Corridor road contract, to construct the $812 million South Road Superway project in South Australia.
Macmahon's share of revenue from the contract is expected to be $230 million. The project will start immediately and is scheduled for completion in 2013.
The Urban Superway Joint Venture team includes John Holland and South Australian company Leed Civil & Engineering, with the contract being undertaken for the Department of Transport, Energy and Infrastructure.
The joint venture will deliver a new 4.8 kilometre highway corridor, including 2.8 kilometres of elevated roadway, connecting the Port River Expressway to Regency Road north of Adelaide. The project is South Australia's largest ever investment in a road project.
