The JV, known as City East Alliance, will undertake the design and building of the highway upgrade, between Kooyong Road and Tonkin Highway.
Stage 1 of the project, over 2.7 kilometres from Kooyong Road to Hardey Road is valued at $225 million.
Work on detailed design is expected to start later this month.
Construction work which will including upgrading all major intersections and include bus priority lanes is scheduled to start in mid-2011 and be completed by the end or 2013.
"This is an extremely important project for the State's economy, given Perth's population growth and the dramatic increase in activity surrounding the Perth Airport precinct," said Transport Minister Simon O'Brien.

The project to upgrade Great Eastern Highway is quickly moving ahead, with the contract being awarded to City East Alliance.
Transport Minister Simon O'Brien said the jointly-funded State/Commonwealth project had been awarded to the joint venture of Leighton Contractors; NRW; and GHD as consultant, to undertake the design and building of the highway upgrade, between Kooyong Road and Tonkin Highway.
Stage 1 of the project, over 2.7km from Kooyong Road to Hardey Road, is valued at $225million. The Commonwealth has indicated further funding would be available under a shared funding arrangement with the State to complete the project through to Tonkin Highway, a distance of 1.4km.
"This is an extremely important project for the State's economy, given Perth's population growth and the dramatic increase in activity surrounding the Perth Airport precinct," Mr O'Brien said.
"It also aligns with the Perth Gateway Project, which is a major upgrade of arterial road network near Perth Airport and the Kewdale freight.
"There will be six lanes on 4.1km of the highway with a central median, on-road cycle facilities and a continuous pedestrian path.
"As part of the works, all major intersections will be upgraded to include bus priority lanes and dedicated turning movements."
Main Roads WA will manage the project under an Alliance contract agreement to ensure the design and building components of the project can be more integrated.
Detailed design is expected to start later this month, with works scheduled to start in mid-2011. The project is expected to be completed by the end of 2013.
