The $220 million stage one contract is for the design and construction of the hospital, where the total capital cost was pegged at $1.76 billion several years ago.
The contract will see Brookfield Multiplex progress the hospital design to a point where major subcontracts for the construction works would be tendered.
The government said that those tender prices will then establish a maximum sum for construction, which if accepted, will result in the company being awarded the stage two contract mid next year.
"Selected 'forward works' would be carried out during Stage One, with the bulk of construction subject to acceptance of the Stage Two contract offer," Health Minister Kim Hames said.
Site works for the 643-bed hospital have been completed in preparation for construction of the main hospital buildings to begin in mid this year. Completion is expected in late 2013.

Property giant Brookfield Multiplex has been appointed as the managing contractor for the Stage One contract for design and construction of the $1.76billion Fiona Stanley Hospital at Murdoch.
Health Minister Dr Kim Hames said Brookfield Multiplex was well positioned to lead the design and construction of the new tertiary hospital - the biggest building project ever undertaken by the State Government.
"Brookfield Multiplex has a reputation for delivering quality, innovative and successful major infrastructure projects, including many within the health sector," Dr Hames said.
Brookfield Multiplex's experience includes the construction of 25 hospitals including:
- Greater Peterborough Hospital, United Kingdom
- Casey Hospital, Berwick, Victoria
- Broadmeadows Health Service, Broadmeadows, Victoria
- Royal Melbourne Hospital, Parkville, Victoria
- Prince Charles Hospital redevelopment, Chermside, Queensland
- Royal Brisbane Hospital, Herston, Queensland
- Waitekere Hospital, New Zealand.
Locally, Brookfield Multiplex has worked on the St John of God Hospital Murdoch and the Queen Elizabeth II Medical Centre, along with the $195million District Court Building, the $225million Perth Convention Exhibition Centre and the $600million City Square development in Perth's CBD.
The Minister said the appointment of the project's managing contractor was another major milestone in the delivery of the new southern metropolitan tertiary hospital.
"The hospital design is well progressed, and site clearing and bulk earthworks are complete in readiness for the start of construction of the main hospital buildings within the coming months," he said.
Brookfield Multiplex Regional Managing Director - Constructions John Flecker said the company was pleased to be involved in the hospital's development.
"It's not often that you have the opportunity to manage the design and construction of a major tertiary hospital from the ground up," Mr Flecker said.
"Fiona Stanley Hospital is set to become one of the best hospitals in the country - we're looking forward to being a part of that."

When completed, Fiona Stanley Hospital would provide comprehensive health care services to communities south of Perth and across Western Australia.
The 643-bed hospital would offer services including:
- a full range of acute medical and surgical services
- State centres for treatment of major trauma, burns, and heart and lung transplants
- WA's first comprehensive cancer service south of the river including radiotherapy treatment facilities, medical oncology and haematology
- renal dialysis
- a major 24-hour emergency department
- paediatrics
- cardiothoracic services
- radiology services including CT and MRI scanning; and
- a state-of-the-art medical research facility to be built in conjunction with universities and the WA Institute of Medical Research.
